A huge plus. In fact, years after taking a new job, one of the interviewing partners showed me that he still had his. It was kind of embarrassing, as I immediately wondered what I had written. But at the same time, it demonstrated that there are people who appreciate the gesture. And for those who don't, I certainly don't think it would work against the applicant. Better to take a few minutes and write one, in my opinion, than to wonder if that seemingly trivial omission was the reason you didn't get a callback.
